I am currently running 35inch Nitto TerraGrapplers (all terrains) on my jk. does the size of the tire effect the reading that the little mileage gauge gives me?

Which gauge are talking about? The odometer or the MPG gauge? Either or the answer is yes. Bigger tires will effect the MPG negatively and the bigger tires will also cause the odometer to be inaccurate. It will read too little mileage.

Buy an aev procal when you get your tires and you can calibrate for the larger tire size

Changing tire size will affect the odometer, speedometer and trip computer (DTE, etc). Unlike in decades past, it takes a software change, versus a change in the speedo gear, to correct it. Various companies (Hypertech, AEV, etc) make programmers that take care of this, as well as other things (i.e., fan speed, diffy gearing, etc). Search for threads on programmers to learn the advantages/disadvantages/costs of each type of device.
