I have a 2007 JK Sahara Unlimited with a manual transmission. I put the rough country 2.5" lift on and now am looking for some 35" tires. It is my daily driver that will probably never go offroad. Will I need to regear?

Ehhhh.....You might be able to get by with just a good tuner. It really comes down to what kind of driver you are. If you're always in a hurry and like to pass people, then yes. However if you're a leave 10 mins early kinda guy then you'll be find. However you'll still need the tuner to fix your speedo.

3.21 gears currently? Will probably want to regear. A tuner or AEV Procal will probably help. The procal is only $150.

Not something you will NEED to do before hand or right when you get your wheels though. You can pretty much try it out and decide for yourself. If you don't like the way it runs, you can research gears and then get them.

If you're not going to go off road, may want to consider 33". Obviously up to you if you want 35's.
