I think I know the answer but double checking just in case.

I will be putting larger tires on, not sure if they will be 33's or 35's, have 3.73 gears and a 6spd manual. Will speedo need to be calibrated?

For sure and if you end up with 35's now or later. You will need gears and a few other items that seem to be discussed on a daily basis here.

You don't really need to calibrate the spedo with 33's. It should only be off by 1 or 2 MPH. I have 33's, it's off by 1 mph. I'm considering an AEV Procal, but mostly to change the TPMS and correct the transmission a bit.

35's will throw it off more, you will probably want an AEV Procal. Not to mention you may want new gears. You don't have to get the gears right away though. Just try it out and decide for yourself.
