Will upgrade locker require readjusting ring-pinion?
If I chose to keep my stock ring/pinion set and just upgrade the locker
inside differential. Do I need to readjust the ring/pinion set after the new locker installed?

The 2 crucial items here ring gear backlash and carrier bearing preload. Check your ring gear backlash before the old diff is removed and copy it when you install the new diff. No pinion adjustment is necessary as you are reusing your gears. Bearing preload is a bit more complicated but still fairly easy.
yes. even though things should go right back together without any change, more times than not, there will be. so long as you can do with dynatrac just said, you should be fine but, the question is whether or not you know how to do it.

Are you sure, from what I read, even intalling a lunch box locker requires moving the carrier
out of differential house, replace the stuff inside the carrier and reinstall carrier back. Since
ring gear is bolted on carrier, this to me means some shift will happen between pinion and ring
