I'm looking to put a set of 33 by 1250 R17 onto a 17 by 9 rim with a negative 12 offset and a 4.5 backspacing will that work on the Jeep JK Unlimited without any rubbing

Yes, I ran that same set up for the past 2 years. Mickey Thompson Baja ATZ 285/75 R17 on Fuel 17X9 wheels with -12 offset and 4.5 BS with bone stock suspension. The front tires rubbed the plastic air damn at full turn. I just removed it. Gas mileage on my 14 JK went from 20 to 16.5.

Yup, that is pretty much the wheel you want to run for up to a 35" tire.
