Noticeable difference between a 3.73 & a 3.21?
It'll definitely cost you to make the swap with a dealer. I would check at some of the local shops and I would certainly go to at least 4.10s or 4.56s if you are planning on adding larger tires. It'll save you the hassle and cost of regearing twice. You'll probably want bigger tires more than once! It's a Jeepers kryptonite.

Where would the gear ratio I have now hurt me the most?
Right now it crawls along pretty well in 4 low in first gear

If you're looking to go 35s, imagine sticking a bicycle in 10th gear and trying to pedal from a stand still. It can be done, but you have to put forth a heck of a lot of effort. It'll be first, extremely noticeable in 1st gear. And then, you will notice through 2-6th, but it's not unmanageable IMO. Don't expect to get into 6th gear as early as before, and especially not uphill! The one risk that you run is if you go wheeling and you are trying to conquer an obstacle... the gears are working that much harder, so breakages may result. Now, if you don't wheel often, you can swing it for a good while before you need a regear. Less stress on the drivetrain.
