I'm completely new to jeeps but the fact that you are towing a trailer would make me want to go to 4.56.

6SPD JKU loaded with 35" tires. - 4.56 definitely. I dare you to go drive an auto for a day. They regularly run at over 3K RPM before shifting. Our 3.6 motors are actually more like a car motor than a truck, its just gearing that makes it a truck application. The 3.6 doesn't even make power until 1800-2200 RPM. I put 4.56 in my 2dr 2012 sport and its great even on the freeways.

I don't know why you say that.
My '12 auto JKU is relatively heavy, with steel bumpers, winch, rock rails, skid plates, and all the recovery gear, camping gear, etc', and it certainly does NOT "regularly run at over 3K RPM before shifting".
That's with 35"s and 4.1 gearing.

If you kick the pedal down to the floor, it is SUPPOSED to shift at higher rpm, to give you the acceleration you want.

High rpm during acceleration isn't a issue at all... But constant high rpm all the time scares me.. 2500-2600 all the time everytime I drive it...

My 2012 with 33's and 6-speed with 3:73's I'm seeing 2800-3000 all day I'm at 7000' elevation I never see 6th gear. I have driven 2 different 2012 JKU's one with 4:56 and the other with 4:88's I'm going with 4:88's with 35's this spring. I'm loaded with armor too its just a dog at this elevation with the hills and wind.

I have a 2013 manual 2 door on 35s. 4.56 gets my 18-19 mpg on the computer, 17s in real time math. I like it, but I have no idea what a 4.10 is like.

Go with the 4.56's. I have 4.10's there is plenty of power but for me, the that six speed clutch is suspect. 4.56 is what I would do and it should be enough for 37's, if you should ever choose to go that high in tire size.

As 4.1 gives plenty of power, and, on the other hand, "4.56 should be enough for 37's", then 4.1 is perfect for the OP, who isn't going to go beyond 35"s...

Had the 456 gears in my 09 jk 2 door. 6 speed. 35 inch tires. The only negative I could see is that first gear was too low to start off in, would have to quickly shift to second. But if I started off in second, had to almost slip the clutch a bit to get rolling. Not enough to hurt the clutch, but just felt a little off. My new 14 is auto with 373 gears. 35 tires. Just added a tuner and it seems fine.
