Vent in tailgate?
A. I rotate my spare in so everything is the same.
B. I picked up a nail last week. I didn't need the spare because of on-board air. I just had to top off and get to Big-O to have it pulled and plugged. BUT a spare is 1) in most states legally required and 2) there for when it's needed. I go by the thought - better to have it and not need it than to need it and not have it.

Think of the air as a solid occupying all of the otherwise unoccupied space in the Jeep. When you close the door and push more air in, with the arc of the door, the air inside needs to go somewhere. Just like water, it will flow in the direction of least resistance - which in this case is any crack or opening like the vent.
