When you can hold speeds without downshifts, use OD.

When you need more rpm's, turn it off.

42RLE—Automatic, Four-Speed Overdrive
Gear Ratios 1st 2.84
Gear Ratios 2nd 1.57
Gear Ratios 3rd 1.0 <--OD Off
Gear Ratios 4th 0.69 <--OD
Gear Ratios Reverse 2.21

Pro: changes shift points and smoother shift in acceleration. Helps keep a higher rpm
Cons: if driving highway at 2300 rpm could eat up some fuel.

Yes.

If you are off-road, off. If you are on a flat freeway, on. If you are going up a steep hill, off. If you are driving around town, on. If you see it shifting in and out of overdrive, off….

Any place with a 3% or higher grade, turn OD off. This is assuming the stock 3.73's. The 3.21's I am really sorry if you live around mountains and haven't re-geared. My buddies rubi 4.1 is driveable in the Northwest with stock BFG's, 4.10's, and OD on.

Get a SuperChips Flashpaq and you probably won't have to worry about it anymore....

The OD off button is there for specific situations such as towing or off-road use where it is desirable to hold 3rd gear. It's good for engine braking or preventing the transmission from hunting between 3rd and 4th when climbing a long incline as well.

Otherwise just leave it on for best fuel economy.
